Summary:

SPERA PHARMA, Inc., in collaboration with Rigaku, has successfully determined the crystal structure of inseparable polymorphic mixtures using cutting-edge 3D Electron Diffraction (3D ED / MicroED) technology.

3D ED / MicroED leverages a transmission electron microscope (TEM) to observe numerous nanocrystals and selectively irradiate targeted crystals with an electron beam. This advanced approach enables structures determination of mixtures that were previously considered impossible using conventional X-ray diffraction methods.

In this study, we tracked an exceptionally challenging case: a mixture of Casopitant mesylate Form 1 and Form 3ーthese polymorphs are inseparable and exhibit nearly identical powder X-ray diffraction (PXRD) patterns. Our team achieved rapid and accurate structure determination, making a significant breakthrough in crystallographic analysis for drug development.

This achievement opens new possibilities for analyzing crystalline powders and complex mixtures that cannot be resolved using traditional X-ray techniques.
